The Roman Colosseum is not a "half-hour" visit: the ticket includes three sites (Colosseum, Roman Forum and Palatine) that make up one of the largest archaeological areas in the world. Getting your timing right is key to neither rushing nor wrecking your day's schedule.

If you only care about the amphitheatre, one hour to an hour and a half is plenty to walk the tiers, see the main level, look down over the Arena and the underground from above and take the classic photos. That is how long most visitors who go on their own with the audio-guide ticket take.
Here is the most common mistake: people book the Colosseum and forget that the same ticket gives access to the Roman Forum and the Palatine, two huge spaces that deserve time. To see it all at a relaxed pace, allow between 3 and 4 hours. As the ticket is valid for 24 hours, a good strategy is to see the Colosseum one afternoon and leave the Forum and Palatine for the next morning, when there are fewer people.
The tour with access to the Arena lasts between 2.5 and 3 hours and includes the guide's explanations, the walk through the Colosseum, entry to the Arena platform where the gladiators fought and, usually, the Forum and the Palatine. Afterwards you can stay on your own for as long as you like within the 24 hours of validity.
